Group I Metabotropic Glutamate Receptors are composed of two receptor proteins, mGlu1 and mGlu5, which are coupled to PLC and intracellular calcium signalling.  These receptors are found in forebrain and cerebellum.
To date, evidence has emerged indicating that group I metabotropic glutamate receptor (mGluR1 and mGluR5) antagonists have antidepressant-like activity in a variety of preclinical models.
Although evidence now exists that these receptors are potential drug targets for ischemia, schizophrenia, and epilepsy, the clinical applications of agonists and antagonists are only beginning.
