Axons are guided along specific pathways by attractive and repulsive cues during the establishment of the nervous system network. Netrin is one these cues. Netrin is a chemoattractant to axon growth cones through the receptors DCC and UNC-5. However, after axonal growth cones cross the nervous system midline, they change their responsiveness to midline guidance cues: they become repelled by the repellent Slit and simultaneously lose responsiveness to the attractant netrin. This is large due to the activation of the Slit receptor Roundabout silences the attractive effect of netrin-1.
